The use of LC-MS-based methods for the quantification of biopharmaceuticals (macromolecular drugs) in biological samples has grown considerably over the past few years. In this presentation, an overview will be given of some recent technical and conceptual developments in the area of biopharmaceutical LC-MS. On the one hand, attention will be focused on improvements in methodology, such as sample handling, separation and detection; on the other hand, the impact of selecting a particular approach on the meaning of the concentration results will be discussed. Several preclinical and clinical case studies of a variety of biopharmaceuticals and biomarkers will be used as illustrations.
