Presentation Description: Multiplex immunoassays have gained lot of momentum recently given the insight one can gain by monitoring an entire pathway or cascade of biological events indicating the efficacy of investigational new drug. Several vendors offer a variety of multiplex kits with different analytes at varying sensitivities and dynamic ranges. While these commercially available kits have the biggest advantage of spitting out tremendous amount of information with limited sample volume, their biggest limitation is the significant variability between lot to lot and performance of critical reagents. The quantitative range and sensitivities of the several MSD V-plex kits we have tested in-house changed significantly between lot-to-lot. Hence, this has become a serious issue during bioanalysis which can last for over five years or more depending on the clinical trial. Since it is not advisable to change the quantitative range within a given study, here we propose some solutions to mitigate this issue.
