This talk will begin with an overview of ASDs- need of such formulations, their design, advantages, disadvantages. This will be followed by understanding their dissolution behavior-how drug and polymer impact the release of ASDs, phase transformations during dissolution, role of dissolution medium on performance and why it is not possible to attain optimal performance at high drug loading. Different strategies to increase drug loading without compromising dissolution will then be discussed. These strategies involve- 1) exploiting ionization behavior of the drug to enhance drug loading in the ASD. 2) incorporating additives to improve dissolution of ASDs. In this strategy impact of different types of additives on the dissolution and subsequent precipitation/crystallization behavior of a model compound will be discussed. Additives selected in this study will cover hydrophobic, hydrophilic excipients and surfactants. Finally, the talk will be concluded by summarizing the observations and how this topic has addressed the need of the industry in the area of developing high drug load ASD formulations.
