We describe a calibration method we call "modified standard addition" that is applicable to the analysis of biomarkers and other endogenous analytes. The method should reduce the need for surrogate matrices or analytes. We provide two example studies, evaluate the calibration method's performance relative to surrogate matrix methods, and discuss limitations and considerations of the calibration method so that others may appropriately apply the modified standard addition method when needed.
