According to International Diabetes Federation 2017, about 425 million people worldwide are suffering from Diabetic Retinopathy (DR). DR is one of the main complications in diabetes patients which damage the retinal blood vessel and lead to blindness.
The present work is an attempt to reposition 5-Fluorouracil (5-FU) which exhibits antifibrotic, anticancer, and anti-angiogenic activities. Hence, topical Nanostructured Lipid carriers (NLCs) of 5-FU were prepared to improve its retinal bioavailability by crossing the blood retinal barrier (BRB).
Developed formulation was optimized by quality by design (QbD) and evaluated for particle size and its distribution, zeta potential, entrapment efficiency (EE). The optimized formulation exhibit nano size range, narrow size distribution, and good EE. The findings are encouraging to further explore this delivery system for in vitro and in vivo studies. Hence, 5-FU loaded NLCs offer a delivery tool for topical treatment of ocular diseases in the posterior segment of eye.
