The maximal drug load is often the problem when wanting to use lipid-based drug delivery systems (LbDDS) for oral delivery of poorly soluble drugs. To overcome this challenges several formulation approaches have recently been identified. These include supersaturation of the drug in the LbDDS, modifying the composition of the LbDDS, including addition of polymers, and also forming lipophilic complexes with the drug prior to inclusion in the LbDDs. These approached have been studies using in vitro lipolysis models and also in vivo pharmacokinetic studies in rats, in order to identify the optimal pathway to load a drug into a LbDDS.
