The use of dried blood volumetric absorptive microsampling (VAMS) for pharmacokinetic assessment of investigational drugs offers many advantages over traditional sampling collection. However, obtaining a complete sample of whole blood onto the VAMS tip can be challenging. The Tasso Hemolink device adds an automated capability to collect blood samples into VAMS cartridge from locations other than the fingertips (e.g. arm) to allow simple, intuitive and virtually painless self-collection of blood samples. The marriage of this technology coupled with liquid chromatography-tandem mass spectrometry (LC/MS/MS) analysis was evaluated for Gefapixant a P2X3 antagonist as a therapeutic agent for chronic cough.
The dried blood VAMS assay was fully validated, and proven robust via extended stability assessments, and very agreeable incurred sample repeatability results. The blood to plasma ratio calculated from the dried blood VAMS and plasma study data was in good agreement with previously determined in vitro assessment.
