In this presentation, we describe the use of thaw-and-use pre-labeled target cells for ADCC assays. The target cells were pre-labeled with calcein AM, cryopreserved in single-use aliquots and used directly in assays after thawing. The thaw-and-use cells pre-labeled with calcein AM showed viability similar to unlabeled cells, superior label retention and slower spontaneous release compared to freshly labeled cells. Further, thaw-and-use pre-labeled cells behaved comparably to freshly labeled cells in ADCC assays involving various effector cells including PBMCs and engineered NK cells, and they were stable for at least 3 years. Most importantly, ADCC assays employing both thaw and-use effector cells and target cells pre-labeled with calcein AM showed improved assay precision and robustness. This novel approach is advantageous because it offers shorter assay times, a reduction in labor and reagents, flexible assay scheduling, and more consistent assay results.
