MP1-17 - Positive correlations between the intensity of histopathological Ubiquitin-conjugating enzyme E2O staining with progression of prostate cancer

Introduction & Objective :
The tumor suppressive functions of AMPK that include inactivation of the mTOR signaling pathway, downregulation of the glycolytic pathway to exert an anti-Warburg effect, arrest of the cell cycle in concert with the stabilization of p53 and p27Kip1 and opposition of the epithelial-mesenchymal transition associated with tumor invasion and metastasis. Ubiquitin-conjugating enzyme E2O (UBE2O), which specifically targets AMPK for ubiquitination and degradation, is known to be amplified in human cancers. This study was performed to evaluate positive correlations between the intensity of immunohistochemical UBE2O staining with progression factors in patients with prostate cancer.
Age, Gleason score, volume (%), size of tumor, bilaterality, zone, perineural invasion, location (apex or base), seminal vesicle invasion, high grade prostatic intraepithelial neoplasm, lymph node invasion, plasma PSA level, clinical stage were examined. Tissue microarrays (TMA) of human prostate cancer (n=180) and control (n=22) were examined. TMA slides were incubated with antibodies against UBE2O, a biotin-conjugated secondary antibody and then incubated with an avidinbiotin-peroxidase complex. Visualization was performed using 3-amino-9-ethylcarbazole chromogen. The TMA cores were scored by the pathologist blind to cancer outcomes. According to histological scoring, the intensity of staining was ranked into one of four groups: high (+++), medium (++), low (+), and negative (-).
Results : In immunohistochemical analysis of human prostate tissue, UBE2O (-) displayed normal prostate histology whereas UBE2O +/+ consistently exhibited prostate cancer. The intensity of UBE2O staining increased in prostate cancer patients with high PSA, Gleason’s score, clinical stage compared with those with low PSA, Gleason’s score, clinical stage of prostate cancer. Importantly, a high degree of positive correlations between the intensity of UBE2O staining and high PSA, Gleason’s score, clinical stage of prostate cancer. It confirmed that the clinical significance of the intensity of UBE2O staining through a cancer patient with high PSA, Gleason’s score, clinical stage of prostate cancer.
Conclusions : In prostate cancer, the intensity of UBE2O staining positively correlates with high PSA, Gleason’s score, clinical stage of prostate cancer. Therefore, the prognosis of prostate cancer can be evaluated with the intensity of UBE2O staining and pharmacological blockade of UBE2O can inhibit the progression of prostate cancer.
