Multiplex Gene Expression and Genotyping using BHQ Probes on the automated IntelliQube

Utilizing Array Tape® technology, the IntelliQube® integrates liquid handling, thermal cycling, detection, and data analysis into one real-time quantitative PCR instrument. The presentation will describe the use of the IntelliQube in conjunction with BHQ® probes for gene expression analysis using one-step RT-PCR. We will also highlight the use of BHQplus probes for duplex SNP genotyping using fast thermal cycling protocols. RT-PCR performance will be analyzed for consistency across singleplex and multiplex formats. In the SNP genotyping experiments, accuracy and reproducibility of this platform were assessed using purified gDNA samples from cell lines obtained from the NIGMS Human Genetic Cell Repository at the Coriell Institute for Medical Research. Assay performance will be compared between standard and fast thermal cycling conditions, with fast cycling capable of producing over 9,200 data points per day with a 45-cycle protocol. Genotype results will also be compared to previously published literature.

Niko Tsatsos

Niko Tsatsos, Ph. D. is an Application Scientist at LGC where he facilitates the scientific consultation, assay optimization and data verification for customers transitioning to a new platform. Dr. Tsatsos received his Ph.D. from the University of Minnesota in Biochemistry and Molecular Biology. His research included gene expression and signaling related to lipid metabolism in mammals. Previously, Dr. Tsatsos worked in the IVD industry developing new clinical assays for DiaSorin and Beckman Coulter. Tsatsos also served as an adjunct Professor at the University of St. Thomas in Molecular Biology during his graduate studies. Dr. Tsatsos joined LGC in 2014 and currently works closely with customers, to help meet the application needs in various industries from Ag-Biotech (breeding/traceability testing), to the clinical market (Pharmacogenomics, SNP analysis, CNV, gene expression etc.) using LGC’s automated solutions
