Traditional single readout assays can provide deep insight into biological systems - especially when couple with high throughput screening methodologies. Multiplexed readout assays such as e.g. two-color assays are a logical extension that furthers our insight into biological systems but have inherent issues in complex biological systems of heterogeneous manner such as patient derived cell lines or primary material. Here, multi-modal readouts have the potential to provide insights in to mode of action of drugs and biological phenomena alike. In the first part of this talk we will highlight how multimodal readouts can be utilized to provide insight into how interferon receptor signaling regulates checkpoint receptors PDL-1 and PDL-1 in patient derived melanoma cell lines using functional genomics. In the second part we will demonstrate how chemical genomics can be combined with force phenotyping to yield insights into differences between traditional readouts such as calcium flux measurements which are thought to play a major role in cell contractility vs. actual measurements of such contractility based on imaging of live patient cells on soft patterned cell substrate.
