Cell death is important for multicellular homeostasis but it also has an unfortunate role in human diseases. Discovery of targeted cytotoxic and cytoprotective agents is vital for treating cancer or neurodegeneration. In order characterize the dysregulation in a disease model, and responses to experimental stimuli, it is important to define the mechanism of cell death and response kinetics. Traditional experimental approaches are often time consuming, labor intensive, costly and inadequate. Here we describe Promega’s new RealTime-Glo™ Annexin V Apoptosis and Necrosis Assay that provides the simplicity of an add-mix-measure assay, in a live-cell, real-time format. While the CLARIOstar with atmospheric control unit(ACU) provides the sensitive detection of duplexed luminescent and fluorescent signals it maintains appropriate [O2] and [CO2] environment for your cells. This combination allows walking away once the test is set up and ensures that you will not miss important cellular responses regardless of when changes happen. Using K562 cells we explored the effects of drug treatment and exposure to natural killer cells on the apoptotic response to uncover time- and dose-dependent responses.
