1357-C - Protein dialysis with automated liquid handling devices

Protein sample preparation is one of the most often tasks in the lab work. Desalting, rebuffering, renaturation, precipitation, resolving of proteins are necessary to purify and to characterize the molecules of interest within samples. The most gentle method under preservation and restorage of protein activity is the dialysis of protein samples. The classical dialysis tools like dialysis tubes or cassettes are not suitable for automated handling with liquid handling devices.  scienova has developed dialysis tools in combination with Deep Well Microplates suitable for automated liquid handling. Examples for the excellent volume recoveries of scienova dialyzer ED300, MD100, MD1000 with STARlet Hamilton Robotics device and Grid Kit with the liquide handling device CyBio Felix of Analytik Jena will be given. Guanidin Hydrochloride is is one of the most often used compounds in concentrations of 6M to dissolve proteins from inclusion bodies. In the next step is it necessary to reduce t he high Guanidin Hydrochloride concentration of the protein sample in order to renature the unfolded protein chains.  A forwarding method to determine exactly the Guanidin Hydrochloride concentration into the sample suitable for automated liquid handling will be shown.
