A high throughput 1536 well phenotypic cell-based assay has been developed using the Cisbio HTRF phospho-IRF3 kit, utilizing FRET based energy transfer between donor and acceptor. Several cell types and plate formats were tested during assay development. THP-1 cells transduced by virus were ultimately selected for HTS and screening was completed with GSK full diversity collection comprising roughly two million compounds. The assay performed well with an average signal to background of 4 and robust Z’ of 0.7. Assay development and screening methods will be described in this poster
