1074-E - Membrane‐free culture and real‐time barrier integrity assessment of perfused intestinal epithelium tubes

In vitro models that better reflect in vivo epithelial barrier (patho-) physiology are urgently required to predict adverse drug effects. Here, we introduce extracellular matrix-supported intestinal tubules in perfused microfluidic devices, exhibiting tissue polarization and transporter expression. Forty leak-tight tubules are cultured in parallel on a single plate, and their response to pharmacological stimuli is recorded over 125 hours using automated imaging techniques. A study comprising 357 gut tubes is performed, of which 93% are leak-tight before exposure. EC50‐time curves could be extracted that provide insight in both concentration and exposure time response. Full compatibility with standard equipment and user‐friendly operation makes this the first Organ-on-a-Chip platform that is readily applicable in routine laboratories.

Remko van Vught, PhD is the team leader of business development at MIMETAS. He is a biochemist by training and obtained his Master’s degree in Molecular Life Sciences cum laude at the Radboud University Nijmegen (2009). During his studies he worked on the structure-function relation of MRP4/ABCC4 at Radboud Institute for Molecular Life Sciences (Nijmegen, the Netherlands). At Pepscan Therapeutics (Lelystad, the Netherlands) he studied the targeting of LRP5 to increase bone formation in osteoporosis (TALOS, FP7). He received his PhD at the Utrecht University (Netherlands) where he worked on the development of functionalized nanobodies (antibody fragments) for cancer therapy (2014). After his PhD research, he started as Business Developer at MIMETAS and is responsible for all business development and marketing activities.
