Dysregulation in iron homeostasis is detrimental because iron is an essential element required for many biological activities in human body. High levels of iron in the peritoneal cavity during the menstruation have been implicated in the pathogenesis of endometriosis. However, the effects of iron in endometriotic cells were poorly characterized. In this study, we investigated the effect of iron (FeSO4 and Ferric ammonium citrate) on cell growth, the production of intracellular ROS (reactive oxygen species), and the expression of inflammatory genes in human endometriotic cells (12Z). Iron did not induce any significant change in the growth of 12Z cells. In contrast, treatment with iron increased intracellular levels of ROS in 12Z cells in a dose-dependent-manner. In addition, iron treatment induced the mRNA expression of inflammatory factors (COX-2, MCP-1, IL-6, TNF-α). Taken together, these results suggest that iron may be associated with the pathogenesis of endometriosis by regulating the production of intracellular ROS and expression of inflammatory in endometriotic cells. Further studies should be performed to confirm the exact effect of iron and its molecular mechanism of action in endometriotic cells.
