P08-101 - Effect of Smallanthus sonchifolius roots (Ss) on the prevention of glucose and lipid metabolism disorders in mice fed a high fructose diet (HF).

The increase in excess gain weight worldwide has closely related with the occurrence of several metabolic disorders as insulin resistance, diabetes type II, dyslipidemia and fatty liver disease. A high fructose diet (HF) might play a role in the onset of those metabolic disorders.The tuberous yacon root is rich in bioactive substances. It contains high levels of fructooligosaccharides (FOS) and also some inulin and phenolic compounds. Some studies have showed that Ss has antioxidant, hypoglycemic and hypolipidemic properties. For most of the studies using Ss, the root powder was obtained using classic tchniques of extraction such as maceration followed by organic extraction and evaporation. In this study, our Ss extract was obtained using a spray drying encapsulation technique with the intuit to prevent as much as possible the bioactive loss. The aim of this work was evaluate the atomized Smallanthus sonchifolius extract to prevent glucose and lipid metabolism disorders in mice fed a high fructose diet (HF).
Swiss male mice (n = 6 / group) were fed during 8 weeks with: balanced diet + water (C), balanced diet enriched with 5% of Ss + water (CS), balanced diet + 30% of fructose on water (HF) and balanced diet enriched with 5% of Ss + 30% of fructose on water (HFS). Food (g) and water (ml) were measured daily. Body weight (g) was measured weekly. After 8 weeks, biochemical measurements (mg/dl) such as glucose (GLU), total cholesterol (CHO), high density cholesterol (HDL) and tryglicerides (TG) were evaluated in blood using commercial kits. Liver lipid content, liver histology and glucose tolerance test (GTT) were also performed. Results were expressed as mean ± SEM. Data were analyzed using t-test. p<0.05 was considered significant for all data. It was observed in HF mice an increase in adipose tissue depots, GLU, CTO, HDL, liver lipid content and also and impaired on GTT and liver histology. On the other hand, HFS mice showed a decrease in liver lipid content, GLU, and also improvement in GTT, liver lipid content and histology.
It seems that Smallanthus sonchifolius was able to attenuate glucose and lipid metabolism alterations in mice fed a HF diet. It is possible that more pronounced results could be achieved with increase the dose and also the period of Ss administration.
