Pterostilbene (PTS) is a naturally occurred phytochemical found in blueberries and grapes. It has many health benefits such as anti-inflammation, anti-cancer and anti-ageing. There are many studies reported the biological activities of PTS. However, there was still a lack of potent detection method for the intracellular trafficking of PTS inside biological tissues. The purpose of this study was to develop a fluorescent labeling method that will enable the visualization of PTS and investigate its preliminary application. Dansyl chloride (DNSC) was used to prepare pterostilbene fluorescent conjugate (PTS-DNSC). The conjugate was formed by refluxing for 2 hours and further purified on a silicon column. The PTS-DNSC has a yellowish color under ambient light and fluorescence under UV light. The conjugate has the excitation wavelength peak at 405 nm and emission wavelength peak at 534 nm. Mass spectroscopy analysis indicated the conjugate was formed between hydroxyl group on pterostilbene and sulfonamide group on Dansyl chloride. Cell study showed this conjugate has similar toxicity at low dose (93% vs 92% cell viability at 10μM ) and less toxicity at high dose (80% vs 46% cell viability at 100 μM). PTS-DNSC was detected in Caco-2 cell using a fluorescent microscopy. In conclusion, a fluorescent conjugate was synthesized with potential application in detecting pterostilbene in biological tissue and understand the internalization mechanisms.
