Background: Biomarkers of cranberry product intake in human are currently lacking. They are needed to verify the compliance of participants in cranberry related clinical trials. The compliance is unknown in majority of trials but is presumed to be low due to the astringency of cranberries. Metabolomics research helps to identify intake biomarkers and provides missing pieces of a puzzle to explain the physiological effects of cranberries. Objectives: To investigate plasma metabolome changes after 3 days and 21 days cranberry juice consumption in a double blinded, placebo-controlled, cross-over intervention study and compare discriminant metabolites with a previous 3-day open-labelled study.
Methods: A double blinded, placebo-controlled, cross-over intervention study were conducted in 17 healthy young women aging 18-29 with normal BMI. Plasma samples were collected at baseline, after 3 days, and 21 days consumption of double strength cranberry juice or a placebo juice. Global metabolomic analysis of plasma samples were conducted using UHPLC-Q-Orbitrap-HRMS.
Results: Separation of two treatment groups were observed after both 3 days and 21 days juice consumption on OPLS-DA (orthogonal partial least squares discriminant analysis) models. One subject was removed from data analysis as an outlier. In positive ionization, 103 discriminant metabolites were found in S-plot after 3 days consumption, 19 of them were observed increasing after cranberry or apple juice consumption in a previous open-label study. Ninety discriminant metabolites were found after 21 days consumption, 17 of them were observed increasing after cranberry or apple juice consumption in the previous data. In negative ionization, S-plot revealed 112 discriminant metabolites after 3 days consumption, 18 of them were observed previously. One hundred and three discriminant metabolites were found after 21 days consumption, 17 of them were observed previously.
Conclusion: Alteration of plasma metabolome was observed among participants after 3 days and 21 days consumption. Multivariate analysis verified discriminant metabolites found in a previous open-labeled study and revealed additional discriminant metabolites as candidates of cranberry intake biomarkers.
