The aim of this study is to evaluate filling quality depending on canal obturation techniques using glucose leakage model, fluild infiltration technique and confocal micropscopy. Forty-six premolars were used in this study (IRB approval no: 2-2015-0086) and divided into three experimental groups (n=10): 1) Continuous wave technique with gutta-percha and AH Plus, 2) Continuous wave technique with BC point and EndoSequence BC sealer, and 3) Single cone obturation technique with BC point and BC sealer. After canal filling, each specimen was subjected to glucose leakage test for nine weeks. At nine week period, fluid infiltration rate of all samples was measured by nano-Flow (IB system, Seoul, Korea) and the selected samples were observed using confocal microscopy. The amount of leakage during nine weeks in each experimental group was statistically analyzed by linear mixed model using SAS(ver.9.2, SAS Inc. Cary, NC, USA) (P<.05). There was no statistically significant difference in the amount glucose leakage and fluid infiltration rate among the three experimental groups. There was no significant correlation between the leakage amount and flow rate. Confocal images showed leakage around the sealer, root canal wall and dentinal tubules in all the samples. This study demonstrated that single cone obturation technique with EndeSequence BC sealer exhibits similar sealing efficacy as continuous wave technique when glucose leakage test model and fluid infiltration measurement were used.
