PR79 - Antibacterial Action of Copper Nanoparticles (CuNPs) on an Ex Vivo Model of Multiespecies Anaerobic Biofilm

Introduction: From an anatomical point of view, the root canal system is complex because in micrometric dentinal tubules, the biofilm is established and its persistence could lead to endodontic failure. Copper nanoparticles (CuNPs) have antimicrobial properties against different microorganisms and could be an alternative for disinfection during endodontic treatments. Aim: To evaluate the antibacterial activity of CuNPs compared to Cu nanowires and ZnO nanoparticles in an ex vivo anaerobic biofilm endodontic model. Methods: Using confocal microscopy analysis with LIFE/DEAD® Baclight™ Bacterial Viability Kit. The optical sections of the biofilm structure were recorded and analyzed using IMARIS software 7.5.2. (Kruskal Wallis. p value<0.05).  Results: There were significant differences in red/green intensities between CuNPs and other nanostructures. The red intensity, corresponding to non-viable bacteria, was significantly higher. There was a statistically significant reduction when exposing the biofilm for 10 minutes at a concentration of 250 μg/mL of CuNPs as for the total biomass (p=0.0001). Exposing the biofilm for two minutes at a concentration of 1000 μg/mL CuNPs, statistically reduced the total biomass (p=0.0018) compared to the negative control, but not with the rest of the nanostructures tested. Conclusion: A better antibacterial activity of CuNPs was observed on a multispecies anaerobic endodontic biofilm. Comparing with other nanostructures, a reduction of viability and biomass was detected when the biofilm was exposed for 10 minutes to a concentration of 250 μg/mL of CuNPs. Further biocompatibility CuNPs studies on PDL cells are needed to support its potential use in endodontics.
