PR63 - The Effect of Lunasin on VEGF release from Activated Macrophages

The anti-inflammatory properties of Lunasin, a soy peptide, has been associated with reduction of inflammatory mediators through suppression of NF-kB pathway. Vascular endothelial growth factor (VEGF) is a heparin-binding protein with specific affinity to endothelial cells and plays a key role in angiogenesis. Previous studies have reported that VEGF could be directly or indirectly regulated by NF-ƘB pathway. No study to date has investigated the effect of Lunasin on the release of VEGF. Thus, this study aimed to investigate the effect of Lunasin on VEGF release from activated macrophages. We hypothesized that Lunasin could significantly increase the level of VEGF release from activated macrophages. THP-1 cell lines that resemble human macrophages were used. After exposure to phorbol-12-myristate-13-acetate(PMA), THP-1 cells differentiated into macrophages. Cell lines were then divided into eight different groups of activated macrophages that were treated for 24 hours with three different concentrations (25 μM, 50 μM and 100 μM) of Lunasin and inactive Lunasin, LPS (positive control) and no treatment (negative control). Levels of VEGF were measured using ELISA. Data was analyzed using ANOVA and post hoc Tukey's test (α=0.05). Regardless of the concentration, all Lunasin treatment groups significantly increased the VEGF release from activated macrophages, compared to positive control, negative control, and inactive Lunasin groups (pConsidering the increased levels of VEGF following the addition of Lunasin, its adjunctive role in the revascularization protocol may show great promise in the outcome of regenerative endodontics.
